چکیده

In this study, we applied bacterial artificial chromosome (BAC) technology with PRVΔTK/gE/gI as the base material to replace first, central, and terminal segments of US3 gene codon-deoptimized fragments via two-step Red-mediated recombination in E. coli GS1783 cells. The three constructed BACs were co-transfected gI part gE carrying homologous sequences (gI+gE’), respectively, swine testicular These recombinant viruses codon de-optimization ((PRVΔTK&gE-US3deop−1, PRVΔTK&gE-US3deop−2, PRVΔTK&gE-US3deop−3) obtained purified. exhibited similar growth kinetics parental AH02LA strain, stably retained deletion TK fragments, inherited recoded US3. Mice inoculated intraperitoneally or control virus PRVΔTK&gEAH02 at a 107.0 TCID50 dose. immunized PRVΔTK&gE-US3deop−1 did not develop clinical signs had decreased load attenuated pathological changes lungs brain compared group. Moreover, mice challenged 100 LD50 provided protection that PRVΔTK&gEAH02. Finally, was injected intramuscularly into 1-day-old PRV-negative piglets dose 106.0 TCID50. Immunized showed only slight temperature reactions mild signs. However, high levels seroneutralizing antibody produced 14 21 days post-immunization. addition, immunization 105.0 complete prevented shedding by 106.5 PRV variant 1 week post immunization. Together, these findings suggest displays great potential vaccine candidate.

Role of pseudorabies virus Us3 protein kinase during neuronal infection.

The pseudorabies virus (PRV) Us3 gene is conserved among the alphaherpesviruses and encodes a serine/threonine protein kinase that is not required for growth in standard cell lines. In this report, we used a compartmented culture system to investigate the role of PRV Us3 in viral replication in neurons, in spread from neurons to PK15 cells, and in axon-mediated spread of infection. We also exam...

Identification of Synonymous Codon Usage Bias in the Pseudorabies Virus UL31 Gene

Background: Little knowledge of synonymous codon usage pattern of pseudorabies virus (PRV) genome, especially the UL31 gene in the process for its evolution is available. Objectives: In the present study, the codon usage bias between PRV UL31 sequence and the UL31-like sequences was identified. Materials and Methods: We used a comprehensive analysi...

Pseudorabies virus US3 protein kinase mediates actin stress fiber breakdown.

Disruption of specific components of the host cytoskeleton has been reported for several viruses and is thought to be beneficial for viral replication and spread. Our previous work demonstrated that infection of swine kidney (SK-6) cells with pseudorabies virus (PRV), a swine alphaherpesvirus, induced actin stress fiber breakdown. In the present study, using several PRV deletion mutants, we fou...

The attenuated pseudorabies virus strain Bartha fails to package the tegument proteins Us3 and VP22.

The Bartha strain of pseudorabies virus has several recognized mutations, including a deletion in the unique short region encompassing the glycoprotein I (gI), gE, Us9, and Us2 genes and point mutations in the gC, gM, and UL21 genes. We have determined that Bartha has mutations in the serine/threonine kinase encoded by the Us3 gene relative to the wild-type Becker strain. Our analysis revealed ...

Pseudorabies Virus

Etiology • Pseudorabies virus (PRV) is an enveloped, double-stranded DNA virus belonging to the family Herpesviridae. Also known as suid herpesvirus-1 (SuHV-1), the virus causes Aujeszky’s disease. • ‘Classical’ PRV strains affecting multiple species were first isolated in the early 1900s. ‘Variant’ PRV strains emerged in swine in China in 2011. • Genomic sequencing and phylogenetic analyses ha...
